Do glucosamine and chondroitin affect MMP13?

The lab evidence is thin and mixed. Glucosamine lowered MMP13 in one study of human cartilage cells and stem cells, but in another it didn't change cytokine-driven MMP13. Other studies found glucosamine lowered MMP-3 and chondroitin lowered MMP-2 and MMP-9. None of these studies measured MMP13 in people.

Glucosamine: one study for, one against

Derfoul and colleagues grew human stem cells and cartilage cells from normal and damaged joints as small pellets, some stimulated with IL-1 beta, and added glucosamine at different doses (Derfoul 2007). At 100 micromolar, glucosamine raised type II collagen and aggrecan gene activity. It partly blocked IL-1 beta's suppression of those genes and inhibited MMP13 expression in both cell types. High doses, 10 to 20 millimolar, worked against it (Derfoul 2007).

Imagawa and colleagues grew human cartilage cells from hip surgery for five weeks with IL-1 beta and oncostatin M, with or without 2 mM N-acetyl glucosamine (Imagawa 2011). Glucosamine cut the cytokine-driven rise in IL-1 beta fourfold. But MMP13 rose with the cytokines whether or not glucosamine was there, and the methylation of its gene promoter didn't change (Imagawa 2011).

Different forms, doses and set-ups gave different answers.

Glucosamine and MMP-3

Dodge and Jimenez treated cartilage cells from patients' knees with glucosamine sulphate at 0.2 to 200 micromolar (Dodge 2003). Aggrecan protein rose with dose, reaching 120% at 150 micromolar. MMP-3, a sister enzyme of MMP13, fell by 18 to 65%, along with its activity. Cells from 40% of the patients didn't respond at all (Dodge 2003).

MMP-3 isn't MMP13, though the two often move together in the studies here. Curcumin with omega-3 lowered both in human cartilage cells, and vitamin C stopped MMP-1, MMP-3 and MMP13 rising in a cartilage tumour cell line (Jhun 2025; Chiu 2016). A fall in MMP-3 hints at a change in MMP13 but doesn't show one.

Chondroitin

Hsu and colleagues treated a human cartilage cell line, chon-001, with chondroitin sulphate (Hsu 2022). It raised type II collagen and the MMP inhibitors TIMP-1 and TIMP-2, and lowered the levels and activity of MMP-9 and MMP-2. It also reduced activation of the Akt and NF-kB pathways (Hsu 2022). The abstract doesn't report MMP13 and doesn't give the concentration.

No study on this site tests chondroitin directly against MMP13.

Why the doses are hard to read

These studies used glucosamine at concentrations from 0.2 micromolar to 20 millimolar in a dish (Dodge 2003; Derfoul 2007), and the higher end blocked the benefits (Derfoul 2007). A dish isn't a joint, and none of these abstracts links its concentrations to an amount taken by mouth.

Where that leaves the question

For glucosamine there is one human cell study showing MMP13 going down and one showing no effect on MMP13 (Derfoul 2007; Imagawa 2011). For chondroitin there is evidence on other MMPs and on TIMPs, not on MMP13 (Hsu 2022). Both are building blocks of the cartilage matrix. Glucosamine is a constituent of the glycosaminoglycans in cartilage (Derfoul 2007), and chondroitin sulphate is part of the matrix of connective tissue (Hsu 2022). Whether either changes MMP13 in a person hasn't been tested in the studies here.

Other questions people ask

Are glucosamine and chondroitin the same thing?

No. Glucosamine is an amino sugar found in cartilage glycosaminoglycans; chondroitin sulphate is a larger molecule in the matrix of connective tissue. They have been tested in separate studies.

Does the form of glucosamine matter?

The studies used different forms: glucosamine sulphate, N-acetyl glucosamine and unspecified glucosamine, at different doses, and their results differed. They weren't compared head to head.

Has anyone measured MMP13 in people taking them?

Not in any study on this site. All of the MMP findings come from cells in a dish.
